Anticoagulant and factor Xa-like activities of Tityus discrepans scorpion venom Acta Toxicol. Argent.
Brazón,Josmary; Guerrero,Belsy; Suze,D; Sevcik, Carlos,Gina; Arocha-Piñango,Carmen L.
Tityus discrepans venom (TdV) produces a variety of haemostatic manifestations including alveoli fbrin deposition and/ or prothrombin and partial thromboplastin time (PT, PTT) alterations in mammals. In vitro studies have demonstrated that TdV contains tissue plasminogen activator-like (t-PA), fbrinolytic and plasmin inhibitory compounds and produces platelets activation through GPVI and a novel Src-dependent signalling pathway. The aim of this study is to describe the initial characterization of procoagulant and anticoagulant components from TdV. This venom was fractionated by exclusion molecular chromatography on a Sephadex G-50 column. The eluted material was collected as fve fractions called S1 to S5. These fractions and the whole venom were used to...

Bone Marrow Mesenchymal Stem Cells Regulate Coagulation and Inflammation Together in Methotrexate Induced Lung Injury Rat Model BABT
Haggag,Nawal Zakaria; El-Shinnawy,Nashwa Ahmed; Abd-Elhalem,Sahar Sobhy; Rashed,Laila Ahmed.
Abstract Clinical research has shed the light on the relation between coagulation and inflammation. Coagulation cascade is activated in lung injury resulting in thrombotic and fibrotic lesions. Such a cascade is initiated by inflammation, then the two systems intense each other. New therapies that modulate coagulation and inflammation will be more successful than therapies targeting only one of them. Mesenchymal stem cells showed anti-inflammatory functions in animal models. The role of mesenchymal stem cells in methotrexate induced lung injury model was evaluated, but no studies scoped on the role of stem cells in coagulation associated with inflammation in such models. This study focuses on the therapeutic role of mesenchymal stem cells against the...

Bothrops alternatus ENVENOMING IN YOUNG DOGS J. Venom. Anim. Toxins
PEREZ,O. A. DE; KOSCINCZUK,P.; FLINTA,S. M.; MAIDANA,H. R.; SANCHEZ NEGRETTE,M..
Bothrops alternatus venom was intramuscularly inoculated (3 mg/kg) into 12 dogs, 30 to 65 days old. Spontaneous bleeding commenced twenty minutes later. Blood samples obtained 3 and 20 minutes after venom inoculation presented spontaneous clotting formation. Plasmatic fibrinogen decreased within 3 minutes. Partial thromboplastin time (PTT) and one-stage prothrombin time (PT) were found. Plasma did not coagulate 40 minutes after inoculation. Platelet counts did not vary but their function was altered. Histopathology pointed out severe muscular necrosis and massive hemorrhage in the inoculation area. Regional ganglia showed intense hemorrhage. The 45 and 65-day-old animals showed alveolar thickening of the septum and generalized congestion, but the...

Evaluation of hemostatic changes using thromboelastography after crystalloid or colloid fluid administration during major orthopedic surgery BJMBR
Topçu,I.; Çivi,M.; Öztürk,T.; Keleş,G.T.; Çoban,S.; Yentür,E.A.; Okçu,G..
The effects of Ringer lactate, 6% hydroxyethyl starch (HES) (130/0.4) or 4% succinylated gelatin solutions on perioperative coagulability were measured by thromboelastography (TEG). Seventy-five patients (ASA I-III) who were to undergo major orthopedic procedures performed under epidural anesthesia were included in the study. Patients were randomly divided into three groups of 25 each for the administration of maintenance fluids: group RL (Ringer lactate), group HES (6% HES 130/0.4), and group JEL (4% gelofusine solution). Blood samples were obtained during the perioperative period before epidural anesthesia (t1, baseline), at the end of the surgery (t2), and 24 h after the operation (t3). TEG data, reaction time (R), coagulation time (K), angle value (α),...

Functional and biological insights of rCollinein-1, a recombinant serine protease from Crotalus durissus collilineatus J. Venom. Anim. Toxins incl. Trop. Dis.
Boldrini-França,Johara; Pinheiro-Junior,Ernesto Lopes; Arantes,Eliane Candiani.
ABSTRACT Background: The prevalent class of snake venom serine proteases (SVSP) in Viperidae venoms is the thrombin-like enzymes, which, similarly to human thrombin, convert fibrinogen into insoluble fibrin monomers. However, thrombin-like serine proteases differ from thrombin by being unable to activate factor XIII, thus leading to the formation of loose clots and fibrinogen consumption. We report the functional and biological characterization of a recombinant thrombin-like serine protease from Crotalus durissus collilineatus, named rCollinein-1. Methods: Heterologous expression of rCollinein-1 was performed in Pichia pastoris system according to a previously standardized protocol, with some modifications. rCollinein-1 was purified from the culture...

Influence of the apparent molecular size of humic substances on the efficiency of coagulation using Fenton's reagent Anais da ABC (AABC)
DE JULIO,MARCELO; DE JULIO,TATIANE S.; DI BERNARDO,LUIZ.
This work used Fenton's reagent as a coagulating agent in the treatment of water samples with high true colour caused by humic substances (HS) extracted from peat. In addition, the effects of the apparent molecular size of HS on coagulation, flocculation and flotation were studied. To that end, four distinct water samples having the same true colour were prepared using HS with different molecular sizes, which were obtained by ultrafiltration fractioning. Through optimisation of coagulant dosage and coagulation pH, as well as posterior construction of coagulation diagrams for each water sample, it was verified that the sample prepared with the smallest apparent molecular size of HS was the most difficult to treat, requiring higher coagulant (Fenton's...

Normal range values for thromboelastography in healthy adult volunteers BJMBR
Scarpelini,S.; Rhind,S.G.; Nascimento,B.; Tien,H.; Shek,P.N.; Peng,H.T.; Huang,H.; Pinto,R.; Speers,V.; Reis,M.; Rizoli,S.B..
Thromboelastography (TEG®) provides a functional evaluation of coagulation. It has characteristics of an ideal coagulation test for trauma, but is not frequently used, partially due to lack of both standardized techniques and normal values. We determined normal values for our population, compared them to those of the manufacturer and evaluated the effect of gender, age, blood type, and ethnicity. The technique was standardized using citrated blood, kaolin and was performed on a Haemoscope 5000 device. Volunteers were interviewed and excluded if pregnant, on anticoagulants or having a bleeding disorder. The TEG® parameters analyzed were R, K, α, MA, LY30, and coagulation index. All volunteers outside the manufacturer’s normal range underwent extensive...

Novel role of antiplatelet agents (aspirin plus clopidogrel) in an incoagulable blood of a victim of russell's viper snakebite J. Venom. Anim. Toxins incl. Trop. Dis.
Bawaskar,H. S..
Snake antivenom is a specific antidote to the venom action, neutralizing the circulating venom. However, it fails to neutralize the venom fixed to target organs such as platelets, renal tubules, etc. Russell's viper venom initiates rapid coagulation in a victim by activating blood platelets, factors V, X, and anticoagulant cofactors. Activation of thrombin, resulting in formation of micro-thrombi, fibrinolysis, and a vicious cascade, sets in. Inhibition of activated platelets by aspirin (cyclooxygenase inhibitor) and clopidogrel (ADP receptor inhibitor) helps to break this vicious circle induced by Russell's venom and may initiate the natural physiological clotting mechanism. They can be utilized as an adjuvant treatment.
